Silicon dioxide is generally recognized as safe by the FDA. In the pharmaceutical industry, colloidal silicon dioxide is used in tablet-making, including use as an anti-caking agent, adsorbent, disintegrant, or glidant to allow powder to flow freely when tablets are processed.
Fumed silica also known as pyrogenic silica is a very fine particulate or colloidal form of silicon dioxide. It is prepared by burning SiCl4 in an oxygen-rich hydrogen flame to produce a "smoke" of SiO2.
Hydrophilic fumed silica is used with increasing success in high technology fields including pharmaceuticals, polyester, silicone, paints and coatings.
Hydrophobic fumed silica has a low moisture adsorption, excellent dispersibility, and ability to adjust rheological behavior, even that of polar systems.

Specifications of Colloidal Silicon Dioxide USP NF Grade:
SiO2 --- 60.08
(This is a fumed silica grade.)
Colloidal Silicon Dioxide is a submicroscopic fumed silica prepared by the vapor-phase hydrolysis of a silicon compound. When ignited at 1000 for 2 hours, it contains not less than 99.0 percent and not more than 100.5 percent of SiO2.
Packaging and storage: Preserve in well-closed containers.
A: Transfer about 5 mg to a platinum crucible and mix with about 200 mg of anhydrous potassium carbonate. Ignite at a red heat over a burner for about 10 minutes, and cool. Dissolve the melt in 2 mL of freshly distilled water, warming if necessary, and slowly add 2 mL of ammonium molybdate to the solution: a deep yellow color is produced.
B: [Caution: Avoid contact with o-tolidine when performing this test, and conduct the test in a wellventilated hood.] Place 1 drop of the yellow silicomolybdate solution obtained in Identification test A on a filter paper and evaporate the solvent. Add 1 drop of a saturated solution of o-tolidine in glacial acetic acid to reduce the silicomolybdate to molybdenum blue, and place the paper over ammonium hydroxide: a greenish blue spot is produced.
pH: between 3.5 and 5.5, in a 1 in 25 dispersion.
Loss on drying: Dry it in a tared platinum crucible at 105 for 2 hours: it loses not more than 2.5% of its weight. Retain the dried specimen, in the crucible, for the test for Loss on ignition.
Loss on ignition: Ignite the portion of Colloidal Silicon Dioxide, retained from the test for Loss on drying, at 1000C ± 25C to constant weight: the previously dried Colloidal Silicon Dioxide loses not more than 2.0% of its weight.
Arsenic: To pass the test. The limit is 8 microg per g.
Assay: Transfer about 500 mg of Colloidal Silicon Dioxide to a tared platinum crucible, ignite at 1000C ± 25C for 2 hours, cool in a desiccator, and weigh. Add 3 drops of sulfuric acid and add enough alcohol to just moisten the sample completely. Add 15 mL of hydrofluoric acid, and in a well-ventilated hood evaporate on a hot plate to dryness, using medium heat (95C to 105C) and taking care that the sample does not spatter as dryness is approached. Heat the crucible to a red color with the aid of a Bunsen burner. Ignite the residue at 1000 ± 25 for 30 minutes, cool in a desiccator, and weigh. If a residue remains, repeat the procedure, beginning with “add 15 mL of hydrofluoric acid.” The weight lost by the assay specimen, previously ignited at 1000C± 25C, represents the weight of SiO2 in the portion taken.
Specifications of Hydrophobic Colloidal Anhydrous Silica BP Ph Eur Grade:
Colloidal silicon dioxide partly alkylated for hydrophobation.
Content: 99.0 per cent to 101.0 per cent SiO2 (ignited substance).
Appearance: White or almost white, light, fine, amorphous powder, not wet-able by water.
Solubility: Practically insoluble in water and mineral acids except hydrofluoric acid. It dissolves slowly in hot solutions of alkali hydroxides.
A. About 25 mg ignited in a platinum crucible at 900 ± 50C for 2 h gives the reaction of silicates.
B. Water-dispersible fraction (see Tests).
Chlorides: Maximum 250 ppm.
To 1.0 g add 30 mL of methanol and 20 mL of dilute nitric acid. Heat on a water-bath for 15 min stirring frequently. Cool, dilute to 50 mL with water and filter. Dilute 10 mL of the filtrate to 15 mL with water.
Water-dispersible fraction: Maximum 3.0 per cent.
Place 0.400 g in a 500 mL separating funnel, add 100 mL of water and shake for 1 min. Allow to stand for 1 h. Allow 90 mL of the aqueous phase to run out dropwise without filtration into a suitable dish dried at 140C and cooled in a desiccator. Evaporate to dryness at 140C, starting at a low temperature to avoid splashing. Cool in a desiccator. The residue weighs a maximum of 12 mg.
Loss on ignition: Maximum 6.0 per cent, determined on 0.200 g by ignition in a platinum crucible at 900 ± 50C for 2 h. It is advisable to place the crucible in a cold oven and then to heat up the oven. Allow to cool in a desiccator before weighing.
To the residue obtained in the test for loss on ignition add sufficient ethanol (96 per cent) to moisten the residue completely and 0.2 mL of sulfuric acid. Add 6 mL of hydrofluoric acid and evaporate to dryness on a hot-plate at about 100C, taking care to avoid loss from sputtering. Wash down the sides of the platinum crucible with 6 mL of hydrofluoric acid and evaporate to dryness. Ignite at 900 ± 50C, allow to cool in a desiccator and weigh.
The difference between the mass of the residue obtained in the test for loss on ignition and the mass of the final residue gives the amount of SiO2 in the quantity of the substance to be examined.

Colloidal hydrated silica produced by precipitation or gelation process.
Content: 98.0 per cent to 100.5 per cent of SiO2 (ignited substance).
Appearance: White or almost white, light, fine, amorphous powder.
Solubility: Practically insoluble in water and in mineral acids, with the exception of hydrofluoric acid. It dissolves in hot solutions of alkali hydroxides.
About 20 mg gives the reaction of silicates.
Solution S: To 2.5 g add 50 mL of hydrochloric acid and mix. Heat on a water-bath for 30 min, stirring from time to time. Maintain the original volume by adding dilute hydrochloric acid. Evaporate to dryness. Add to the residue a mixture of 8 mL of dilute hydrochloric acid and 24 mL of water. Heat to boiling and filter under reduced pressure through a sintered-glass filter (16). Wash the residue on the filter with a hot mixture of 3 mL of dilute hydrochloric acid and 9 mL of water. Wash with small quantities of water, combine the filtrate and washings and dilute to 50 mL with water.
pH: 4.0 to 7.0.
Suspend 1.0 g in 30 mL of a 75 g/L solution of potassium chloride.
Water-absorption capacity: In a mortar, triturate 5 g with 5 mL of water, added drop by drop. The mixture remains powdery.
Substances soluble in hydrochloric acid: Maximum 2.0 per cent.
In a platinum dish, evaporate to dryness 10.0 mL of solution S and dry to constant mass at 100-105C. The mass of the residue is not more than 10 mg.
Chlorides: Maximum 0.1 per cent.
Sulfates: Maximum 1 per cent.
Iron: Maximum 300 ppm.
Loss on ignition: Maximum 20.0 per cent, determined on 0.200 g in a platinum crucible by heating at 100-105 °C for 1 h and then at 900 ± 50 °C for 2 h.

Silicon Dioxide occurs as an amorphous substance that shows a noncrystalline pattern when examined by X-ray diffraction. It is produced synthetically, either by a vapor-phase hydrolysisprocess, yielding fumed silica, or by a wet process, yielding precipitated silica, silica gel, colloidal silica, or hydrous silica. Fumed silica is produced in an essentially anhydrous state, whereas the wet-process products are obtained as hydrates or contain surface-adsorbed water.
Fumed silica occurs as a white, fluffy, nongritty powder of extremely fine particle size and is hygroscopic. The wet process silicas occur as white, fluffy powders or as white, microcellular beads or granules and are hygroscopic or absorb moisture from the air in varying amounts. All these forms of Silicon Dioxide are insoluble in water and in organic solvents but they are soluble in hydrofluoric acid and in hot, concentrated solutions of alkalies.
Function: Anticaking agent; defoaming agent; carrier; conditioning agent; chillproofing agent in malt beverages; filter aid.
A. Place about 5 mg of sample into a platinum crucible, mix with 200 mg of anhydrous potassium carbonate and ignite over a burner at a red heat for about 10 min. Lead: Not more than 5 mg/kg.
Loss on Drying: Fumed Silica: Not more than 2.5%; Precipitated Silica and Silica Gel: Not more than 7.0%; Hydrous Silica: Not more than 70.0%; Colloidal Silica: Not more than 85.0%.
